Cadmium affects glutathione-related metabolism in Vicia faba roots

چکیده
Cadmium (Cd) is one of the most toxic heavy metals in plants. To investigate whether Cd induces common plant defense pathways, different physiological and molecular reactions, including modifications in glutathione (GSH) level, activity and gene expression of glutathione reductase (GR) enzyme were characterized in roots of hydroponically cultivated-Vicia seedlings during 12, 24 and 48 h. Vicia roots which accumulated the metal, showed an increase in GR1-like gene transcripts, encoding for GR enzyme, as earlier as 12 h after the onset of the Cd treatment while the root growth was not as yet affected by the Cd treatment. When Vicia roots exposure was longer (24 h and 48 h), metal induced significant decreases in fresh (FW), dry weight (DW) and water content. These growth changes were accompanied by progressively increases in GR activity, GSH and oxidized glutathione (GSSG) level.

Biomass growth, micronucleus induction, and antioxidant stress enzyme responses in Vicia faba exposed to cadmium in solution.
Biomass growth, micronucleus induction, and antioxidative stress enzymes (superoxide dismutase, peroxidase, glutathione reductase, and catalase) were investigated simultaneously in the Vicia faba plant exposed to cadmium in solution. The biomass lowest-observed-effect concentration (LOEC) value was 2,000 microM Cd2+. In the shoots, enzymic activities increased without concentration-response rel...
متن کاملThe Role of Heavy Metals on Metabolism of Polyamines in Bean Plants (VICIA FABA) and its Susceptibility to Fusarium Wilt
Faba bean plants (Vicia faba) showed remarkable resistant to wilt disease caused by Fusarium solani pv. Phaseoli when watered with a mixture of heavy metal at a concentration 10mg/L. However the use of Zn, Pb alone slightly increases the susceptibility of bean plants to Fusarium wilt. Plants exposed to Cd or Cu treatment accelerate the symptoms appearance in infected plant. The toxicity of a si...
متن کاملFavism: current problems and investigations.
Haemolytic favism is a severe, acute anaemia which occurs in glucose-6-phosphate dehydrogenase deficient individuals, usually following the ingestion of Vicia faba seeds. Current interest is focused on the nature of the active substances of Vicia faba and on the causes of the varying susceptibility among G6PD-deficient individuals to episodes of severe haemolysis. The results of experiments in ...
متن کاملAccumulation of heavy metals and antioxidant responses in Vicia faba plants grown on monometallic contaminated soil
The purpose of this study was to explore the effects of soil contamination by selected metals (cadmium, copper, nickel, lead or zinc) on the antioxidant response of Vicia faba plants. The levels of the antioxidants: glutathione, proline, non-protein thiols, as well as guaiacol peroxidase and catalase activities were measured in the upper parts of plants. Additionally, the potential bioavailabil...
متن کاملSensitivity of Allium cepa and Vicia faba towards cadmium toxicity
Sensitivity of common onion (Allium cepa L.) and faba bean (Vicia faba L.) to cadmium (Cd) stress was investigated using genotoxicity endpoints. Simultaneously, the antioxidative stress enzymes (guaiacol peroxidise and catalase) and lipid peroxidation [malonaldehyde (MDA) content] were investigated in the plants exposed to Cd in solution. The endpoints screened for genotoxicity included chromos...
